In a recent interview with Game Informer magazine, Dan Houser made it clear that Grand Theft Auto 5 will have a "very big map", while also seemingly confirming that Los Santos is the only major city in the game.
Below is a quote from the magazine article, credit to mocha on our forums.
Until we receive any further information however, the facts as they stand are that GTA 5 is set in Los Santos and will feature a very big map.

Game Informer: You just released the GTA V trailer. Can you talk about that project in any respect? Obviously, it looks like it's going back to San Andreas. While this would appear to back up Rockstar's initial press release about the game being set in Los Santos, we still can't call it 100% confirmed. The fact that Dan explained he would "stick to the press release" would appear to show he doesn't want to give anything away that Rockstar aren't ready to discuss - but that there clearly there is more to tell. Whether that is the inclusion of other cities (such as San Fierro and Las Venturas), time will tell.
Dan Houser: I've got to be real careful here, or they will drag me through the office and whip me with barbwire. I will stick to exactly what's in the press release. It's Los Santos and the surrounding countryside - and a very big map.

- Zulu2065 (Guest) June 02, 2012I'm very excited to explore this "very big map" and honestly don't care if SF or LV isn't in here. Just as long as Los Santos is as big as GTA 4 and the country is very big and detailed, I'll be fine.
- GTA 4Life (Guest) May 29, 2012They better put the other two cities in. you can't go back to San Andreas and leave us hanging with one city... no matter how detailed it is. to me, GTA San Andreas is the bench mark. They need to give me that and more
- Mynameisnunya (Guest) March 09, 2012I was sad 'bout not having sf and lv but it depends on The trailers, news and gameplay clips. So far I cease to see anything cause stupid, hawaii tourist, bald, max Payne! I loved the series back in the day but after seeing leukemia tourist, I was disappointed. Just throw mp in the trash and give us gta v already!!!!!
- Akuma (Guest) February 29, 2012hurry up with explaining the map already
- Concerned GTA Fan (Guest) February 28, 2012If they don't include San Ferro then that will be terribly disappointing. That's the only city my brother and I played in. It has all the hills and jumps. :D I fail to see why they made all the other "stories" games and didn't do a GTA San Andreas Stories-That would be giving the public what they want. There was Liberty City Stories and Vice City stories and SA Stories would be the next logical step. I didn't like GTA IV because they totally changed from the fun arcade-like game play that they've carried over from all GTA games since GTA III and made everything too realistic, physic-wise. Ever heard of the saying, "Why tamper with a winning system?" That's why we returned GTA IV and got Saints Row 2 and subsequently got Saints Row The Third. If they made a GTA San Andreas Stories then there'd be no questions about it, your favorite cities would be there along with better graphics and newer cars with the same great gameplay engine. I have a sneaking suspicion that this GTA V will be a disappointment to my bro and I just like IV was... That's why we're going to rent it first. :\
